Most first trips include all three. The question is how to split your nights.
Tokyo
Best for: sheer variety, shopping, nightlife, modern culture, museums. Downsides: huge — travel between areas eats time. Day trips: Kamakura, Nikko, Hakone, Yokohama.
Kyoto
Best for: temples, gardens, traditional architecture and food culture. Downsides: famous sights are crowded and hotels expensive in peak season; buses can be slow. Day trips: Nara, Uji, Kurama, Himeji.
Osaka
Best for: street food, nightlife, value hotels, transport hub for Kansai. Downsides: fewer headline historic sights in the city itself. Day trips: Kyoto, Nara, Kobe, Himeji, Koyasan.

One base for Kansai?
If you dislike changing hotels, base in Osaka or central Kyoto and day-trip to the other — they're 15–30 minutes apart by train.
